Label-free dual assay of DNA sequences and potassium ions using an aptamer probe and a molecular light switch complex.

A homogeneous assay is reported using Ru(phen)(2)(dppz)(2+) and a K(+)-binding aptamer for the selective and sensitive detection of a target oligonucleotide and potassium ions, based on reduction in fluorescence emission according to the formation of the G-quadruplex structure from the aptamer in the presence of K(+).

G-quadruplex-modulated fluorescence detection of potassium in the presence of a 3500-fold excess of sodium ions.

A label-free detection of K(+) was developed using G-quadruplex DNA (c-Myc) modulated fluorescence enhancement of tetrakis-(diisopropylguanidino) zinc phthalocyanine (Zn-DIGP). Upon the addition of increasing concentrations of potassium, a detection limit of 0.8 μM for K(+) was easily achieved. Comparative titrations using sodium, lithium, ammonium, transition metal, or alkali earth salts revea...

Perylene ligand wrapping G-quadruplex DNA for label-free fluorescence potassium recognition.

A perylene ligand, N,N-bis-(1-aminopropyl-3-propylimidazol salt)-3,4,9,10-perylene tetracarboxylic acid diimide ligand (PDI), which consisted of π-conjugated perylene moiety and hydrophilic side chains with positively charged imidazole rings, was used to wrap G-quadruplex for fluorescence turn-on K(+) recognition. Electrostatic attraction between PDI's positively charged imidazole rings and DNA...

Graphene oxide amplified fluorescence anisotropy for label-free detection of potassium ion.

Fluorescence anisotropy (FA) has attracted considerable attention, but it has been rarely applied for the detection of small molecules and metal ions because they are too small to induce evident FA changes. Although some mass amplifying strategies have been developed, the recognition probes need to be covalently modified with the fluorescent dyes, which is complex and expensive. To overcome thi...

A label-free G-quadruplex-based switch-on fluorescence assay for the selective detection of ATP.

A G-quadruplex-based, label-free, switch-on fluorescence detection method has been developed for the selective detection of ATP in aqueous solution using crystal violet as a G-quadruplex-selective probe. The assay is highly simple and rapid, and does not require the use of fluorescent labeling.
